Type □: If checked, the type of every trajectory is determined using their respective properties and recorded in the Results table using the heading TYPE (see Trajectory Properties for more details). Each type consists of specific criteria designed to evaluate trajectories properties. Each trajectory is tested against all the active types until at least one specific criterion is met. If not, the default type (1) is attributed to the trajectory. When a trajectory meets the criterion of a specific type, the associated index, label and color is attributed to the trajectory. When a trajectory meets more than one criterion, the last one is attributed to the trajectory. Press □ to open the Type Options dialog box.
Flag □: If checked, trajectories are analyzed for specific events that could occur along the different positions over time. Press □ to open the Flag Options dialog box.
Motion descriptors +: If checked, Motion measurements including the distance, velocity and confinement ratio are recorded in the Results table (see Trajectory Properties for more details). Uses the heading MOTION in the Results table. Default setting is unchecked.
Type of displacement +: If checked, the type of displacement (Moving, Diffusing and not Moving) is determined and recorded in the Results table (see Trajectory Properties for more details). Uses the heading DISPLACEMENT in the Results table. Default setting is unchecked. This command is enabled when the tracking mode is ‘Mobile’.
Moving: Trajectories with a median value of the velocities measured along the trajectory, higher than the value specified in this field are characterized as ‘Moving’. The value is expressed in pixels or in calibrated distance per frame. Default setting is 2.
Confined: Trajectories with a maximum distance from the origin measured along the trajectory lower than the lower value of the range specified in this field are characterized as ‘Non Moving’. Trajectories with a maximum distance from the origin measured along the trajectory inside the range specified in this field are characterized as ‘Diffusing’. The value is expressed in pixels or in calibrated distance per frame. Default setting is 2.
MSD: If checked, the mean squared displacement is computed on the trajectory. A fit is performed on the MSD and the extrapolated value of the velocity, the diffusion coefficient and the confinement factor are recorder in the Results table (see Trajectory Properties for more details). Note that when the MSD is selected, the extrapolated value of the velocity, the diffusion coefficient and the confinement factor are used to determine the type of displacement. Default setting is unchecked. This command is enabled when the ‘Type of displacement’ command is selected.
Delta: Defines the maximum lag time between two positions. Default setting is unchecked. This command is enabled when the ‘Type of displacement’ command is selected. Value is expressed in ‘frame’. Default setting is 20.
Result(s): If checked, the MSD values as a function of the lag time between two positions are measured along the trajectory and recorded in the Results table (see Trajectory Properties for more details).
Phase detection +: If checked, trajectories are analyzed for specific shift in the velocity, the type of displacement, the direction and any user-defined property that could occur along the different positions over time. When one or more shifts are detected along the trajectory, the segments between two shifts are called ‘phase’. Default setting is unchecked. This command is enabled when the tracking mode is ‘Mobile’.
Lifespan: Selects the minimum lifespan of a phase. Value is expressed in ‘frame’. Default value is 1.
Result(s): If selected, the result of each phase of every trajectory will be displayed in the Results table.
Displacement: If selected, the shift in the direction or in the velocity are analyzed and used to define phases. Default setting is unchecked. This command is enabled when the phase detection is selected.
Direction: If selected, the shift in the direction are analyzed and used to define phases. Default setting is unchecked. The change of direction is detected when the angle formed between the particle on the next frame, the particle on the current frame and the particle on the previous frame, is inside the specified range. Values are expressed in Degrees. Default setting is ‘0-45’. Those commands are enabled when the phase ‘Displacement’ is selected.
Velocity: If selected, the shift in the velocity are analyzed and used to define phases. Default setting is unchecked. The change of velocity is detected when the velocity measured at a specific position passed the moving threshold defined in the ‘Tracking Parameters’. This command is enabled when the phase ‘Displacement’ is selected.
User-defined Property: If selected, the shift in the property defined by user are analyzed and used to define phases. Default setting is unchecked. This command is enabled when the phase detection is selected.
Property: Selects the property to be used. Default setting is empty.
Ratio: The change is detected when the ratio measured between the value of the specified property at the current frame and the value of the specified property at the previous frame is outside the range specific in this field. Default setting is
Filter: Allows any of the 8 moving filters to be selected. When selected, the values associated with the specified property are smoothed using the specified moving filter of the specified size. Default setting is ‘none’.
Filter size: Select the size of the moving filter to be used. The value is expressed in frame. Default setting is 2.
Lineage +: If checked, trajectories are analyzed to reconstruct the lineage upon division.
Mode: Selects the mode to be used to reconstruct the lineage. Two modes are available: Distance and Delta. In the mode ‘Distance’, daughter cells are determined based on the Euclidian distance between the particle center of the mother cell and the center of possible daughter cells. In the mode ‘Delta’, daughter cells are determined based on the range of distances measured on the X and the Y axis, between the particle center of the mother cell and the potential daughter cells.
Distance: The maximum distance between a mother cell and possible daughter cell. The value is expressed in pixels or in calibrated distance. Default setting is 1. This command is enabled when the mode ‘Distance’ is selected.
Delta X: The range of distances between a mother cell and possible daughter cell measured on the X-axis of the image. The value is expressed in pixels or in calibrated distance. Default setting is ‘0-max’. This command is enabled when the mode ‘Delta’ is selected.
Delta Y: The range of distances between a mother cell and possible daughter cell measured on the Y-axis of the image. The value is expressed in pixels or in calibrated distance. Default setting is ‘0-max’. This command is enabled when the mode ‘Delta’ is selected.
Division Result: If selected, the result of each division detected along the different lineages will be displayed in the Results table.
